Islet perifusion involves the <strong>exposure of islets to continuous flow with nutrients, gas, or reagents<\/strong>. Several researchers published perifusion systems with the control of temperature, pH and flow rate of the perifusate.<\/p>\n<p>Currently there is no standardized, inexpensive apparatus and protocol that can be replicated easily in any laboratory.<\/p>\n<\/div>\n<\/div><\/div><\/div><\/div><div id=\"pg-1794-3\"  class=\"panel-grid panel-no-style\"  style=\"margin-bottom: 50px\" ><div id=\"pgc-1794-3-0\"  class=\"panel-grid-cell\" ><div id=\"panel-1794-3-0-0\" class=\"so-panel widget widget_sow-headline panel-first-child panel-last-child\" data-index=\"4\" ><div\n\t\t\t\n\t\t\tclass=\"so-widget-sow-headline so-widget-sow-headline-default-3244f9959cc6-1794\"\n\t\t\t\n\t\t><div class=\"sow-headline-container\">\n\t\t\t\t\t\t\t<h2 class=\"sow-headline\">\n\t\t\t\t\t\tISLETS PERIFUSION\t\t\t\t\t\t<\/h2>\n\t\t\t\t\t\t<\/div>\n<\/div><\/div><\/div><div id=\"pgc-1794-3-1\"  class=\"panel-grid-cell\" ><div id=\"panel-1794-3-1-0\" class=\"so-panel widget widget_sow-editor panel-first-child panel-last-child\" data-index=\"5\" ><div\n\t\t\t\n\t\t\tclass=\"so-widget-sow-editor so-widget-sow-editor-base\"\n\t\t\t\n\t\t>\n<div class=\"siteorigin-widget-tinymce textwidget\">\n\t<p>The <strong>islet perifusion set-up<\/strong> consists of <strong>four perifusion columns<\/strong> connected to four channel Isamtec pumps, to pump 4 different perifusion solution into 4 parallel islets-containing columns simultaneously with the <strong>flow rate of 100 \u00b5L\/min<\/strong>. The LAMBDA OMNICOLL 4-channel sample collector selects the top 4 wells of the 96 well plate every minute <em>(A, B, C &amp; D - each representing a different treatment condition)<\/em> to collect 12 minutes of fractions and then move to the bottom four rows <em>(E, F, G &amp; H rows)<\/em> of the 96-well plate.<\/p>\n<p>The perifusate in the collecting 96-well plates were kept at &lt;4 \u00b0C to preserve the integrity of the analytes.<\/p>\n<blockquote>\n<p><em>LAMBDA OMNICOLL multi-channel fraction collector bases (containing the perifusate samples) can be placed in a cold bath or any other thermo-stabilized container.<\/em><\/p>\n<\/blockquote>\n<p>The collected samples of the perifusate were assayed for <strong>insulin, glucagon, or somatostatin concentration<\/strong>.
